An effecient method for identifing all chromosome pairs of Veronica persica and other rare plants with small size chromosomes.
Although the chromosome number of most Veronica species have been counted, but there have not been seen many works on karyotype analysis of this genus. This matter affects on the analysis and discussions of systematic and phylogenic relationships between species and sub-species negatively.Particularly,detection and identification of sub-species inter-relationships have been accompanied with explicit difficulties.
On the other hand, small size and poor stainability of Veronica chromosomes and many other plants has been caused many problems on the karyotype studies.Therefore,development of a pure and simple method for identification of small chromosomes of Veronica species and plants like this seems to be necessary.On this research we developed a new modified method which was matchable and fitted definitely with Veronica genus.
We tested 30 samples of Veronica persica originated from north of Iran(Gilan province) as a main biological material.The chromosome number of this species was counted 2n=28. Six pairs,numbered V1,V3,V4,V10,V11,V13 were detected such a Metacentic and then 6 pairs,V2,V5,V6,V8,V12 and V14 were identified as a Sub-metacentric.And after all, 2 pairs,V7 and V9 were observed such as Sub-telocentric chromosomes.One pair ,V9 was sattelited chromosome.Application of this method has created a Partial Banding Pattern(PBP) on the whole length of chromosomes.
According to (PBP) pattern the heterochromatic and euchromatic regions were ripping and detaching apart on the length of each chromosome distinctively and individual homologous chromosomes show similar PBP. The whole length of chromosomes were ranged 2.8-6.3 Micron.Also,V2 and V7 pairs were completely heterochromatic and the other chromosomes showed particular PBP.
Other cytologenetical factors like the length of long arm and short arm, arms ratios and the centromeric index also were measured easily.
